Denosumab injection (Prolia) is used to treat osteoporosis (a condition in which the bones become thin and weak and break easily) in women who have undergone menopause (''change of life;'' end of menstrual periods) who have an increased risk for fractures (broken bones) or who cannot take or did not respond to other medication treatments for osteoporosis. to treat men who have an increased risk for fractures (broken bones) or who cannot take or did not respond to other medication treatments for osteoporosis. treat osteoporosis that is caused by corticosteroid medications in men and women who will be taking corticosteroid medications for at least 6 months and have an increased risk for fractures or who cannot take or did not respond to other medication treatments for osteoporosis. to treat bone loss in men who are being treated for prostate cancer with certain medications that cause bone loss, to treat bone loss in women with breast cancer who are receiving certain medications that increase their risk for fractures. Denosumab injection (Xgeva) is used to reduce the risk of fractures in people who have multiple myeloma (cancer that begins in the plasma cells and causes bone damage), and in people who have certain types of cancer that began in another part of the body but has spread to the bones. in adults and some adolescents to treat giant cell tumor of bone (GCTB; a type of bone tumor) that cannot be treated with surgery. to treat high calcium levels that are caused by cancer in people who did not respond to other medications.Denosumab injection is in a class of medications called RANK ligand inhibitors. It works to prevent bone loss by blocking a certain receptor in the body to decrease bone breakdown. It works to treat GCTB by blocking a certain receptor in the tumor cells which slows the tumor growth. It works to treat high calcium levels by decreasing bone breakdown as the breakdown of bones releases calcium.
Xgeva is a monoclonal antibody that is used to treat women with breast cancer that has spread to the bone. It is also used to treat giant cell tumor of the bone, Hypercalcemia of Malignancy, multiple myeloma and other conditions as prescribed by your doctor. It can be used to prevent bone fractures and spinal cord compression in people with multiple myeloma. It is available as a subcutaneous injection.

Injection routeYour doctor will prescribe your exact dose and tell you how often it should be given. This medicine is given as a shot under your skin. It is usually given in your upper arm, upper thigh, or stomach.

This medicine should come with a Medication Guide. Ask your pharmacist for a copy if you do not have one.
This medicine is not right for everyone. You should not receive it if you had an allergic reaction to denosumab, or if you are pregnant.
It is not safe to take this medicine during pregnancy. It could harm an unborn baby. Tell your doctor right away if you become pregnant. If you are a woman who can get pregnant, your doctor may do tests to make sure you are not pregnant before receiving this medicine. Use an effective form of birth control to keep from getting pregnant during treatment with this medicine and for 5 months after the last dose.
Tell your doctor if you are breastfeeding, or if you have kidney disease, diabetes, gum disease, or a history of fractures. Tell your doctor if you have problems with your thyroid, parathyroid, or digestive system.

Do not suddenly stop using this medicine without checking first with your doctor. Doing so may increase your risk for more fractures. Talk to your doctor about other medicines that you can take.
Your doctor will do lab tests at regular visits to check on the effects of this medicine. Keep all appointments.
Pain, swelling, heavy feeling, or numbness in your mouth or jaw, loose teeth or other teeth problems
This medicine can cause jaw problems. You must have regular dental exams while you are being treated with this medicine. Tell your dentist that you are receiving this medicine. Practice good oral hygiene.
Severe bone, joint, or muscle pain
Unusual pain in your thigh, groin, or hip
Do not use Prolia® and Xgeva® together. They contain the same medicine.
Some medicines can affect how denosumab works. Tell your doctor if you are also using medicine that weakens your immune system, including a steroid or cancer medicine.
This medicine can cause jaw problems. You must have regular dental exams while you are being treated with this medicine. Tell your dentist that you are using this medicine. Practice good oral hygiene.

- Muscle spasms or twitching, numbness or tingling in your fingers, toes, or lips
- Pain or burning during urination, change in how much or how often you urinate
- Fever, chills, cough, sore throat, body aches
- Allergic reaction: Itching or hives, swelling in your face or hands, swelling or tingling in your mouth or throat, chest tightness, trouble breathing
- Blistering, peeling, red skin rash
- Chest pain, fast or uneven heartbeat, trouble breathing
- Lightheadedness, dizziness, fainting
- Pain, swelling, heavy feeling, or numbness in your mouth or jaw, loose teeth or other teeth problems
- Unusual pain in your thigh, groin, or hip
- Sudden and severe stomach pain, nausea, vomiting
- Arm or leg pain
- Diarrhea
- Redness, pain, itching, burning, swelling, or a lump under your skin where the shot was given
- Tiredness or weakness
